My PhD project is a collaboration between two laboratories in ACHUCARRO, the laboratory of Neural Stem Cells and Neurogenesis and the laboratory of Neuronal Excitability which aims to set a model of hippocampal organotypic slices to study how seizures induce reactive gliosis and reactive neurogenesis subsequently disrupting synapses and circuitry. The organotypic slice model allows for fast and efficient pharmacological and genetic modifications of signaling pathways. It also allows for high resolution imaging (by 2-photon microscopy) coupled to electrophysiological recordings (from patch clamp to microelectrode arrays). Thus we investigate alterations in different cell types (astrocytes, neurons, neural stem cells) and their impact on the synapse and functional connectivity.
