Role of Galectin-1 in migration in microglia: generation of mutant mESCs by CRISPR Cas9

Abstract

Results previously obtained in our laboratory indicated that Aβ1-42 stimulation increases protein O-GlcNAcylations in microglia. One of the O-GlcNAcylated proteins is galectin-1, which we identified that is being modified by this group in the serine 8. We then found that this post-translational modification in the galectin-1 controls microglial migration. Taking into account that all these experiments were done with the ectopic expression of galectin-1, in order to study the role of O-GlcNAcylation in endogenous galectin-1, we generated galectin-1S8A and galectin-1S8C mESCs by CRISPR Cas9 genome editing technique. Experiments with these cells have shown that the same pattern as in microglia is maintained. In the future, these mutant mESCs will be differentiated into microglia, astrocytes and thyroid cells
